In rodents, the heart possesses the remarkable ability for regeneration within the post-birth. Lactylation is related to the occurrence of heart disease such as heart failure. We determined an integrated study combining global proteomics, lactylome, and genome-wide RNA sequencing to elucidate the role of lactylation throughout postnatal heart development. Proteins in the mouse heart undergo substantial changes in the very early stages after birth and stabilize at 6 weeks and beyond. Lysine lactylation (Kla) plays a role in cardiac development and undergoes dynamic changes throughout the development cycle of the mouse heart, with distinct patterns during early postnatal stages for histones and non-histones. ChIP-seq data indicated that H4K12la plays a role in regulating downstream gene expression in the mouse heart, thereby influencing DNA replication and subsequently affecting the corresponding mouse phenotype. Our results suggest that histone Kla is expected to be a potential target for clinical induction of cardiac regeneration.

As two of the largest economies and countries in the world, the People's Republic of China (China) and the United States of America (U.S.) both have enormous resources and innovative talents in a number of sectors. The U.S.-China Track II Dialogue on Healthcare was set up as a forum to discuss and engage ideas on this very topic. In this dialogue, Dr. Qun Yan, head of thehLifeEditorial Office engages in a dialogue with Professor Mark McClellan, the Director of the Duke-Margolis Institute for Health Policy at Duke University and U.S. Co-Chair of the Track II Dialogue on Healthcare, Professor Gordon G Liu, Professor at Peking University and the Dean of the PKU Institute for Global Health and Development and Co-Organizer of the Track II Dialogue, Mr. Stephen Orlins, President of the National Committee on U.S.-China Relations and a Co-Organizer of the Track II Dialogue, and Professor George Fu Gao, Academician at the Chinese Academy of Sciences and former Director of the Chinese Center for Disease Control and Prevention. They discuss how and why the Track II Dialogue was created, the successes and developments of the dialogues, and what the future of American-Chinese health collaboration looks like.

2.Rethinking BCG vaccine delivery for enhanced efficacy: Are two distinct routes of BCG administration better than one?

In summary, this article highlights the potential of enhancing BCG vaccine efficacy against tuberculosis through alternative administration routes. While intradermal BCG vaccination has long been the standard, its variable effectiveness against adult pulmonary TB suggests the need for new strategies. Recent studies indicate that oral BCG vaccination can significantly boost mucosal immunity, which is critical for protection against respiratory infections. Combining oral and intradermal routes may thus offer a dual advantage, eliciting both systemic and mucosal immune responses. Future research should focus on validating the safety and efficacy of this combined approach in clinical settings to improve TB prevention.

Various clinical trials are under different phases, and in the coming time, drugs might change the landscape of AD treatment.

该研究发现,LRRK2依赖于其激酶活性促进STAT1磷酸化,从而促进巨噬细胞的M1极化以及下游效应分子的表达,对巨噬细胞的免疫功能具有显著作用。这些发现不仅有助于更好地理解Lrrk2在帕金森病发病机制中的作用,也为未来针对免疫系统的治疗策略提供了新的思路——Lrrk2可能成为治疗自身免疫性疾病的潜在靶点。未来,通过进一步研究LRRK2的激酶功能及其在免疫系统中的角色,有望为帕金森病的精准治疗带来新的突破。
