On the cover: The phospholipase D (PLD) enzyme superfamily is pivotal in various cellular processes, notably in oncogenesis. However, its specific role in nasopharyngeal carcinoma (NPC) remains unclear. In this issue, Zhou et al. report the oncogenic role of PLD1 in NPC, demonstrating a positive feedback loop between PLD1 and the NF-κB pathway that drives NPC progression (pp. 997–1006). PLD1 enhances NF-κB activity by promoting the phosphorylation and nuclear translocation of RELA (p65). This nuclear translocation allows RELA to bind to the PLD1 promoter, enhancing its expression. The cover image illustrates this feedback loop, with two mice running on a wheel, symbolizing the mutual reinforcement between PLD1 and NF-κB. The wheel’s mechanical energy is converted into electricity, leading to an overloaded circuit board imprinted with nasopharyngeal tissue, ultimately resulting in system failure due to excessive accumulation.
